Wayne started his professional basketball after being drafted by the Minnesota Timberwolves in the 2009 NBA draft where he was selected as the 28th overall pick. In his first season for the Timberwolves, he recorded an average of 6.6 points per game alongside 2.1 rebounds. He was traded to Memphis Grizzlies in exchange for the player Dante Cunningham after playing for the team for 3 years. On January 7, 2013, Wayne Ellington recorded a new career-high with 26 points in a 113–81 victory against the Sacramento Kings, however, after a few days, he was traded to Cleveland for the forward player Jon Leuer.

Wayne did not stay for quite long with Cleveland as he soon signed with the Dallas Mavericks on July 26, 2013. After one year with the Mavericks, he was traded to Los Angeles Lakers for one season during which he played a total of 65 games and was eventually laid off.
